The winner of the bout on Wednesday earns a place in the 2013 ICC Champions Trophy Final. England has never won the ICC Champions Trophy, going as far as the final in 2004 before losing to the West Indies. South Africa won the inaugural tournament in 1998 but hasn't made it back since, according to Sky Sports.
"It's a massive game for both sides," England captain Alastair Cook told reporters. "We've got the opportunity to go into the final of an ICC competition. It's such an exciting place for a player to be.
"We've almost played knock-out cricket throughout this Champions trophy. This is obviously one step towards the final and we've come here to try and win the tournament."
England will be without spin bowler Graeme Swann, who is struggling with a calf strain. The club has their sights set on the final and doesn't want to risk Swann being unavailable for it. They will also be without Tim Bresnan, who will be out as he awaits the birth of his first child. Bresnan is replaced by Steven Finn.
South Africa could be without the services of paceman Dale Steyn, who has a side strain. Despite playing in front of England's home crowd, South Africa captain AB De Villiers is confident his squad can upset the favorites.
"I believe we can beat them but they're playing in front of their home crowd and I think all the pressure's on them," Villiers told reporters. "We'll make sure we do the basics well like I said before and if things go our way we'll end up on top."
The semifinal match between India and Sri Lanka takes place on Thursday, June 20 at 5:30 a.m. ET.
